Description: pBluescript-FL is created from pBluescript II phagemid SK(+). Its BamHI-SmaI sites (in the MCS) were converted to BamHI-PflMI-SfiI-PflMI sites (SmaI is destroyed in the process). The rest of the vector is untouched. 1st strand cDNA was primed with an oligo(dT) primer. Double-stranded cDNA was ligated into PflMI adaptors and inserted into the two PflMI sites in the T3 (5') to T7 (3') direction. PflMi can be used to isolate the cDNA insert. Library materials provided by G. Elgar (UK MRC HGMP-RC) and constructed and donated by Drs. K. Kawakami, M. Sasaki, S. Sugano, K. Kikuchi, and S. Watabe (University of Tokyo, Institute of Medical Science and Laboratory of Aquatic Molecular Biology and Biotechnology). T3 should be used for sequencing the 5' end of clones and T7 for the 3' end.
